The structural and functional integrity of mitochondria depends on a defined protein and lipid composition of its membranes. Disturbances in the metabolism of cardiolipin, a characteristic lipid of mitochondrial membranes, are associated with various cardiomyopathies including Barth syndrome, Sengers syndrome and dilated cardiomyopathy with ataxia (DCMA). The mitochondrial lipid homeostasis depends on contact sites between both mitochondrial membranes and lipid transfer proteins of the Ups1/PRELI protein family, which mediate the transport of phospholipids between both mitochondrial membranes in a lipid specific manner. Mechanisms of mitochondrial lipid trafficking and its relation to the biogenesis of mitochondrial proteins will be discussed.
